Random transposition of Himar1 in BGC. Genomic DNA was prepared from nine randomly selected kanamycin resistant transformants, digested for 3 h with Not I, and transferred to a nylon membrane. The membrane was hybridized with a probe that detected oriR6K on the transposon. DNA from BGC strain ATCC33664 was included as a negative control (lane -). The positive control (lane P) was pHBurk3. The 7-, 5-, 3- and 2-kb fragments contained in the biotinylated 2-log DNA ladder (New England Bio-Labs) are indicated on the left. The two hybridizing fragments observed in lanes 5 and 7 are indicative of double Himar1 insertions in the respective mutants.
