PLX4032 inhibits incorporation of EdU selectively in mutated papillary thyroid cells. Nthy-ori 3–1 and BCPAP cells were treated with PLX4032 for 36 hours and then exposed to EdU for 1 hour. The incorporated EdU was detected using the “click chemistry” approach , cellular DNA was counterstained with DAPI and intensity of cellular fluorescence was measured by laser scanning cytometry. The data are shown as bivariate (DNA content versus EdU incorporation) scatterplot distributions; insets show DNA content frequency histograms from the respective cultures.
