Effects of miR-155 on JAKs/STATs phosphorylation in IL-6-stimulated RASFs (short time course). RASFs in a density of 5 × 104 cells/well in 12-well culture plates were transfected with miR-155 mimics or control for 48 h, serum-starved (12 h) and stimulated with IL-6 (50 ng/ml) plus sIL-6R (50 ng/ml) for the indicated periods. Phosphorylation of JAK1, JAK2, STAT1, and STAT3 were determined by Western blotting using phospho-specific or pan antibodies against JAK2, STAT1 and STAT3. Phosphorylation of JAK1 was barely detected (data not shown). Three experiments were performed using RASFs isolated from three different RA patients and a representative result is shown
