The larger size of some WHI5 mRNA spots in these experiments may be attributable to the longer time lag between the imaging of two z-stack planes (due to the exposure time of the Hoechst 33342 dye), during which mRNA molecules diffuse, to a higher excitation power, or to the presence of several WHI5 mRNA molecules within the same nuclei.
